Importance of expanding phase II trials when a subgroup analysis has generated a hypothesis
Need to have clear preclinical documentation of a possible MOA and of drug interactions
Demonstration of invivo activity
Importance of knowing tumour genomics
Need to define highly predictive biomarkers that will allow targeting of drugs to specific patients who will derive a clinically meaningful benefit
Need to better understand the interactions between tumour genotype and patient biology.
Innovative and well planned trials, involve oncology experts at early stages of clinical development.
